Activation of Teliospore Germination in Uromyces appendiculatus var. appendiculatus. II. Light and Host Volatiles
نویسندگان
چکیده
The effects of light and host volatiles on teliospore germination in Uromyces appendiculatus var. appendiculatus were studied. I. Light intensities ranging 15-26000 lux supported teliospore germination and basidiospore formation. The optimum light intensity was 17000 lux under the described conditions. Light-dark alternation was necessary for teliosp.ore germination: no germination occurred under continuous light or continuous dark conditions. The process of teliospore germination, basidiospore formation, and basidiospore release followed a synchronous, rhythmic pattern of development. Teliospore germination was triggered by the phase change light -+ dark (i.e. "light off"). Under long-day conditions basidiospore release exhibited a nocturnal periodicity with peak maxima occurring ~ 7 h after light was turned off. 2. Fifteen days after a 10 d exposure to host volatiles, 70-80 % of the treated spores germinated; minor increases in spore mortality were observed. Inoculations made with seedling-treated teliospores resulted in heavy pycnial infections. 3. The heat and seedling activation methods developed for teliospores of bean rust proved ineffective when applied to dormant teliospores from six other unrelated rust species. 1) This study represents a portion of a disser, tation submitted by the senior author to the Faculty of Biology at the University of Konstanz (Fed. The effects of aging and temperature on teliospore germination in Uromyces appendiculatus var. appendiculatus were studied. 1. Indoor storage was good at ~ 4 QC in the dirk. Under these conditions spores remained dormant for ~ 9 mo, but thereafter germination gradually increased to a maximum (~ 63 %) after 36 moo Spores .9tored outdoors showed a marked increase in germination after ~ 4 mo and r...

The cytology of teliospore germination and basidiospore formation in Urornyces appendiculatus vat. appendieulatus was characterized with light and fluoroscence microscopy. Meiosis of the diploid nucleus occurred in the metabasidium. The four haploid daughter nuclei migrated into the basidiospore initials where they underwent a post meiotic mitosis. Each basidiospore was delimited from the meata...

متن کاملProtein accumulation in the germinating Uromyces appendiculatus uredospore.
Uromyces appendiculatus is a rust fungus that causes disease on beans. To understand more about the biology of U. appendiculatus, we have used multidimensional protein identification technology to survey proteins in germinating asexual uredospores and have compared this data with proteins discovered in an inactive spore. The relative concentrations of proteins were estimated by counting the num...
